If I have a very small room (6x10) with an HVAC vent and I place a large object over the top of that vent (not actually laying on the vent, just above it - let's say 1" above the register), will that pose a significant problem to heating/cooling the room? Won't the air just hit the bottom of the object and then pass along the bottom of the object until it finds the open space of the room and then dissipate? Aren't I still delivering the same amount of heat/cooling to the room?

You will restrict it a slight amount as it puts some back pressure on the airflow. Will increase the velocity slightly and sound. Try it and find out if you notice a difference.
